The paper considers the operator \({\mathcal{L}}_0\) in \(L_2(0,1)\) generated by the regular boundary value problem \[ l(u)=u^{(n)}(x)+q_2(x)u^{(n-2)}(x)+\cdots+q_n(x)u(x),\quad 0<x<1, \] \[ U_j(u)=\sum\limits_{k=0}^{n-1}\left[\alpha_{jk}u^{(k)}(0)+\beta_{jk}u^{(k)}(1)\right] =0,\quad j=1,\ldots, n. \] For any fixed integer \(m\in[1,n]\), the paper considers the operator \({\mathcal{L}}_1\) given by \({\mathcal{L}}_0\) and the perturbed boundary condition \[ U_j(u)=0,\quad j=1,\ldots,n,\quad j\neq m, \] \[ U_m(u)=\int_0^1\overline{p(x)}u(x)dx, \quad p\in L_2(0,1). \] The authors construct the characteristic determinant of the spectral problem for \({\mathcal{L}}_1\) under the assumption that the operator \({\mathcal{L}}_0\) possesses a system of eigenfunctions and associated functions generating an unconditional basis in \(L_2(0,1)\). They study the stability and instability of the unconditional basis properties of the system of eigenfunctions and associated functions of the problem under the perturbation. They present the Samarskii-Ionkin problem with integral perturbation of its boundary condition as an example of applications.
