The main objectives of the paper are to give higher-order multiscale asymptotic expansion of the solution for mixed boundary value problems of a second order elliptic equation in a perforated domain, and to present a multiscale finite element method (cf. \textit{J. L. Lions}, [Rocky Mt. J. Math. 10, 125--140 (1980; Zbl 0443.35022)]). The crucial idea of the method proposed in the paper is to use homogeneous Dirichlet boundary conditions on the boundary of the unit cube.
