In this paper, the authors study the tau-function arising from the isomonodromy problem for a general \(\mathrm{sl}(2)\) system with three Fuchsian singularities which can be reduced to the classical Gauss' hypergeometric equation. In particular, they apply the theory of extended monodromy symplectomorphism recently developed by \textit{M. Bertola} and \textit{D. Korotkin} [Commun. Math. Phys. 388, No. 1, 245--290 (2021; Zbl 07424950)] which interprets the Jimbo-Miwa-Ueno tau-function as the generating function on the monodromy map. This approach allows to describe the dependence of the tau-function on the monodromy data and to express explicitly it in terms of the Barnes' \(G\)-function. In addition, the authors show that when the Fuchsian singularities are taken at \(0, 1\) and \(\infty\) the essential part of the tau-function coincides with the structure constant of the asymptotic formula for the solutions of the Painlevé VI equation discovered by Gamayun-Iorgov-Lisovyy.
