Guidelines for the kinetic modeling of endocrine-metabolic systems are discussed. Model validation is of utmost importance nowadays. In this respect concepts of a priori identifiability and optimal experiment design are studied. The theory is illustrated on the glucose-insulin system and on ketone body metabolism.
