The author considers three different types of models in order to interpret BIAcore sensogram data accurately. Inside the BIAcore the evolution is governed by the following equation for the bound state \(\widetilde{B}:\) \[ \frac{\partial \widetilde {B}}{\partial \widetilde{t}} = k_a ( C_T - \widetilde{C}_\Delta) (R - \widetilde{B}) - k_d \widetilde{B}, \] where \(k_a\) and \(k_d\) are, respectively, the association and dissociation rate constants, \(R\) is the initial number of empty receptor sites, and \(\widetilde{C}_\Delta\) is the deviation of the analyte concentration from the inlet concentration \(C_T.\) The three types of models considered are partial differential equation models, an integro-partial differential equation model, and an ordinary differential equation model. These models are used to determine \(\widetilde{C}_\Delta\) accurately. The author then presents analytical results as well as numerical simulations that show the following: for small Damköhler number, the parameter estimates for the ordinary differential equation model are within experimental tolerances when compared with those estimated using the integro-partial differential equation model, but for moderate values of Damköhler number, short-time sensogram data can be generated by two different sets of rate constants for the same model.
