      The goal of this paper is to relate connectedness properties of the spectrum of a ring to local cohomology modules. Second vanishing theorem was proved for local cohomology modules over unramified regular local rings of mixed characteristic, it characterizes the connectedness of punctured spectra through the vanishing of local cohomology. Certain mixed characteristic Lyubeznik number determines the number of connected components of the punctured spectrum of a ring. In last section, the highest mixed characteristic numbers of a collection of rings that includes those with isolated singularities was investigated. It was shown that this number equals the highest Lyubeznik number as they relate to other Lyubeznik numbers, and to connected properties of spectra.
