    The paper focuses on the properties and characteristics of a specific type of derivation in polynomial algebras, say Shamsuddin derivations, and their implications on the isotropy groups and simplicity of these derivations. The author provides an affirmative answer to a conjecture regarding Shamsuddin derivations and their simplicity, namely a Shamsuddin derivation $D$ is simple if and only if \(\mathrm{Aut} (K[x,y_1,y_2,\dots,y_n])_D=\{id\}\). The author also calculates the isotropy groups of these derivations and establishes a connection between the image of a derivation and Mathieu-Zhao subspaces, namely for the Shamsuddin derivations \(D=\partial_x+\sum_{j=1}^r(a(x)y_j+b_j(x))\partial_j\) of \(K[x,y_1,y_2,\dots,y_y]\), the image subspace \(\mathrm{Im} D\) is a Mathieu-Zhao subspace if and only if \(a(x)\) is a constant.
